Abstract : 「緒言」67Ga-citrateは, 悪性腫瘍および急性炎症の病巣の画像診断として広く臨床に利用されている. しかし, 67Gaが細胞に取り込まれるメカニズムは, 現在も完全に解明しておらず, 多くの仮説が提唱されている. 1970年, Hayesらが, 動物腫瘍の実験から, 67Gaの悪性腫瘍への取り込みは, 変性もしくは壊死の組織よりも活動性の組織に多いことを報告した. また, 1972年, Bichelらは, 67Gaの取り込みは腫瘍細胞の増殖と関係があるらしいと報告した. 臨床的には, 1977年と1982年に, Higashiらは, 頭頸部癌および肺癌について, 一般に臨床的な悪性度が強く予後も悪い悪性腫瘍ほど67Gaがよく取り込まれることを報告した. 1980年, Bidaniら, 1981年, 菅原らも同じような臨床例を報告している. 著者は, 67Gaの細胞への取り込みと細胞機能との関係をより詳しく究明する1つの手段として, マウス・白血病細胞とエーリッヒ腹水癌細胞を同調培養して, 細胞周期と67Gaの取り込みの関係を検討した.

Title : The Relationship between Cell Cycle and 67Ga Accumulation in Malignant Tumor Cells

Abstract : [Abstract] : The mechanism of tumor localization of 67Ga is not known with certainty, although much information has been derived regarding the biodistribution and subcellular fate of 67Ga in a variety of tumors and other tissues in experimental animals. Hayes et al. and Ito et al. have reported that uptake of 67Ga in transplanted animal tumors is associated with viable rather than necrotic tissue. In 1982, we reported that increased 67Ga uptake in malignant tumors indicates a more highly malignant tumor. In the present study I have attempted to examine the relationship between cell cycle using synchronized mouse tumor cells and 67Ga accumulation into malignant tumor cells. Cells were synchronized by a modified method of Doida and Okada. 1) The change in mouse leukemic cell number with time after the synchronization procedure showed a typical growth curve of a synchronized population as seen in Figure 3 (A). The incorporation patterns of 14C-thymidine showed a single incorporation peak at 6.5 hours after the end of colcemid treatment as seen in Figure 3 (B). This peak corresponded to the middle of S stage. The incorporation patterns of 67Ga-citrate also showed a single incorporation peak at 5.5 hours after the end of colcemid treatment as seen in Fig 4 (B). This curve corresponded to the 14C-thymidine incorporation curve. 2) The change in mouse Ehrlich's tumor cell number with time after the synchronization procedure showed a typical growth curve of a synchronized population as seen in Figure 5 (A). The incorporation pattern of 67 Ga-citrate showed a single incorporation peak at 12 hours after the end of colcemid treatment as seen in Figure 5 (B). This peak corresponds to the S-G2 transition. Nuclear Feulgen DNA content of mouse Ehrlich's tumor cell was measured with an automatically registering fluorescence cytophotometer. The Feulgen DNA content increased gradually at S stage, the maximum content of Feulgen DNA corresponded to the S-G2 transition as seen in Fig 5 (B). The incorporation pattern of 67Ga corresponded to the pattern of increased Feulgen DNA content. From these results, I conclude that there is correlation between 67Ga uptake and stage of DNA synthesis in malignant tumor cells.
