Abstract : 「緒言」Androgen Target Organの一くに, ラット, マウスの顎下腺が取り上げられ, この顎下腺のAndrogen依存性を扱った報告が多数ある. 我々もこの点についてAndrogenレベルから研究を進めている. そして, ラット顎下腺Microsome中の5α-ReductaseにはTestosteroneに対する親和性に性差が存在すること, 顎下腺内Testosterone, 5α-Dihydrotestosterone濃度は雌雄差なく一定に保持されていること, などを報告し, ラット顎下腺のAndrogen依存性について検索してきた. 今回はさらに, 顎下腺のAndrogen Receptorについて実験を行った. 顎下腺のAndrogen Receptorに関しては, Wilsonら, Dunnら, Verhoevenらのマウス顎下腺での5α-Dihydrotesterone Receptorの報告がある.

Title : R1881 Binding Substance in Cytosol of Rat Submandibular Gland.

Abstract : Present investigations deal with the androgen receptors of cytosol in rat submandibular gland (SMG) using methyltricnolone (R1881) as the labeled ligand. R1881 bound spccifically to cytosol of rat SMG. The sedimentation analysis of the cytosol androgen receptor by sucrose density gradient centrifugation showed a sedimentation rate of 4S. The relative ability of steroids to compete with 3H-R1881 in male rat SMG cytosol was as follows: R1881 > 5α-androstane-3α, 17β-diol (3α-Diol) > triamcinolone acetonidc (TA) > testosterone (T) > estradiol-17β (E2) > dihydrotestosterone (DHT) > progesterone (P4), and in female rat: R1881 > TA > P4 > T > E2 > DHT > 3α-Diol. In rat ventral prostate, the binding of 3H-R1881 was competed by low concentrations of T and DHT. In rat SMG, on the other hand, this binding was competed by high concentrations of T and DHT. From these results it is speculated that the character of R1881 binding substance of cytosol differ in SMG and ventral prostate.
