Abstract : 「緒言」androgen標的臓器であるラット顎下腺では, 組織microsome中に5α-reductaseの活性があること, 雌雄共に同レベルのdihydrotestosterone (DHT) の蓄積が認められ, しかもDHTはtestosteron (T) より多量に存在していることから, 顎下腺での活性型androgenは, DHTであると推測される. さらに活性型androgenとしてのDHTの意義を確立するには, 組織cytosol中のDHTreceptorに関する検索が必要である. DHTのreceptor assayにおける問題点として, (1) Te-BG (testosteron estradiol binding globulin) とligandの結合, (2) ligandとしてのDHTの代謝, (3) 内因性DHTによる影響が挙げられる. これらの問題点を補う物質として, 近年, 合成androgenであるmethyltrienolone (R 1881) が合成され, 前立腺をはじめとするandrogen標的臓器のandrogen receptor assay (ARA) に頻用されている.

Title : A Study on Androgen Receptor in the Rat Submandibular Salivary Gland

Abstract : Three types of 5α-Dihydrotestosterone binding proteins (ABPs) in the rat submandibular salivary gland cytosol from which endogenous steroids were removed by dextran coated charcoal (DCC) method (DCC prepared cytosol) were shown by sucrose density gradient centrifugation (SDGC) and Sephadex G-200 gel filtration chromatography. The sedimentation coefficient of these ABP-adrogen complexes were estimated as approximately 4S, 6S and 9S. And with Sephadex G-200 gel filtration chromatography, 4S, 6S, and 9S DHT binding peaks were eluted nearby the elution regions corresponding to BSA, BGG and void volume, respectively. However, 3H-methyltrienolone (R 1881) binding was not revealed in 6S region・ And in the cytosol which was not treated with DCC, only 4S peak was shown in both 3H-R 1881 and 3H-DHT binding experiments. All these binding peaks were disappeared by adding 1000 fold excess radioinert DHT or R1881. Furthermore, in the submandibular cytosol obtained from 14 days castrated male rats, these binding peaks of ABPs to 3H-R 1881 and 3H-DHT were completely disappcared. Relative ability of various steroids to compete with 3H-DHT binding was as follows: R1881 = DHT > Testosterone >> Estradiol > Progesterone for female, and R1881 = DHT > Testosterone > progesterone > Estradiol for male. These results cleary demonstrated that three types of ABPs (4S, 6S, and 9S) were present in the rat submandibular cytosol and controled by androgens.
