Abstract : 「緒言」 歯科矯正臨床は, 発育過程にある小児を対象に, 咬合の育成に努めることをその目的の1つとしている. 小児の咬合異常の多くは, 顎顔面頭蓋の成長過程に現われるため, 将来生じうる咬合の成長発育に伴う変化を的確に把握するためには, まず周囲組織の成長過程を理解しなければならない. このような観点から, 顎顔面頭蓋の成長発育は, 長い間矯正臨床において極めて重要な研究課題となっている. Hellman (1927) による顔面の成長研究以来, 顎顔面頭蓋の成長発育に関する研究が数多くなされてきた. なかでも頭部X線規格写真を用いた成長研究は, 多くの研究者によっておこなわれてきた. しかしながら, 現在のところ顎顔面頭蓋の成長発育に関する研究は, 成長発育の機序やその調節機構など, 成長発育の本質に迫るところまでは到っていない. これを解明するには, 正常な成長発育の様相を追求するとともに, 成長発育に影響を与えると考えられる遺伝形質, ホルモン, 環境, 成長因子, 代謝などに関する広範な研究が必要とされる.

Title : Effects of the aging and epidermal growth factor on the biosynthesis of macromolecules in rabbit alveolar bone

Abstract : This experiment was designed to understand the metabolic changes of alveolar bone tissue with respect to age and the influence of epidermal growth factor (EGF). Male Japanese white rabbits, 3, 6, 10, 16 and 20 weeks old, were used as experimental animals. The alveolar bones were dissected, and fragmented by means of bone cutter and were then incubated for I20 minutes at 37℃ in Krebs Ringer phosphate buffer, pH 7.4, containing Succinate- 1,4-14C as a tracer with or without 10-9 g/ml of EGF. After the incubation, media and tissues were fractionated into medium non-dializable metabolites, intracellular metabolites, EDTA soluble metabolites and insoluble residues. These were then analyzed for synthesizing macromolccules. Incorporation of radioactivity into subdevided metabolites showed that the synthesis of macromolecules in alveolar bone gradually decreased with aging while EGF was able to activate the production of macromolecules especially in l6 weeks old animals. The macromolecules from mcdium, non-dializable and intracellular fractions were separated into three peaks, peak I, II and III, by Sepharose CL-6B molecular sieve chromatography under dissociating condition (7M urea). Although the incorporation of the radioisotope into each peaks decreased with aging, the relative proportion were not affected by aging. Addition of EGF activated the incorporation of radioactivity into the peaks at about a 3 times greater rate than that of control in 16 weeks old animals. Alveolar bone cells actively produce a hyaluronic acid. EGF increased the propotion of hyaluronic acid twice than of the control.
