Abstract : The morphological changes and the number of mast cells in the submandibular gland of the ovariectomized rats treated with estrogen, and of the testectomized rats treated with androgen were studied. The effects upon the mast cells of both hormones gave similar results; Following administration of these sexual hormones, the number of mast cells also revealed a significant reduction. In addition, a considerable degranulation of mast cells was observed. This result was also supported by electron microscopic observation. By means of alcian blue-safranin staining method, there were many safranin positive mast cells in the controls, while in the estrogen and the androgen treated animals, alcian blue-positive mast cells increased. In specimens treated with both hormones, orcein-water blue positive mast cells were markedly decreased. Quantitative microfluorometry showed that the mast cells in the controls contain large quantities of serotonin. After the administration of both hormones the content of serotonin in the mast cells was prominently reduced. An examination of the tongue of both hormones treatment revealed no corresponding reduction in the number of mast cells. These results suggest that there is a possible biological role of mast cells in the metabolism concerned with the sexual dimorphism of the submandibular gland and their secretion.
