Abstract : 「緒言」 永久歯歯胚の一連の成長過程の変化を石灰化matrix proteinばかりでなく, 非石灰化matrix proteinについても生化学的に解明することは, 小児歯科学領域においても重要な基礎的研究と考えられる. 近年, 生化学領域の発展に伴い, 局所のコラーゲンが加齢変化に対してアミノ酸組成の変化および架橋結合変化についてBaileyら, および高木らによって報告された. さらに, 桜井はウシ永久歯歯胚を用いて歯胚形成過程において, コラーゲンと非コラーゲン成分が加齢と共に変動することを報告した. また, 中山は結合組織の主要な非コラーゲン性物質であるglycosaminoglycansが歯胚形成期と萌出期で明らかに異なることを報告した. 従って, 永久歯歯胚, 特に歯胚歯髄および歯小嚢の主要な有機質成分をなすコラーゲンが歯胚形成にどの様な機能をもっているかを検索する一つの手段として, コラーゲン分子の分子間および分子内にどのような架橋結合が存在するかを明らかにする必要がある.

Abstract : The biochemical study of the unerupted tooth germ in the developmental stages of the tooth is of great value in all areas of dentistry, and especially in pedodontic practice. In this study, collagenous and non-collagenous components of the tooth germ were fractionatcd with neutral salt solution and the cross-links and their intermediates of insoluble reduced collagen were analyzed to investigate the developmental processes of tooth germ. By the extraction of 1.0M NaCl/0.05M Tris-HCl (pH 7.4), it was found that the soluble components of the stage III of dental papilla (or pulp) and dental sac were remarkably decreased and the insoluble components were 98.9+-0.7% and 92.1+-2.6%, respectively. Following reduction with sodium borotritide (NaB3H4), the specific radioactivity of collagen at the every stage of dental papilla (or pulp) was constantly 500 cpm 3H/μg collagen but that of dental sac was increased 40,122 and 306 cpm 3H/μg collagcn at stages I, II and III, respectively. The reduced cross-links, hydroxylysinonorleucine (HLNL) and dihydroxylysinonorleucine (DHLNL) together account for a major portion of the radioactivity comprising up to 70% of the total radioactivity and their relative abundance varied with stages. Moreover, the ratios of DHLNL to HLNL were 1.74-1.94 in dental papilla (or pulp) and 0.56 and 0.52 at stage I and III in dental sac. In contrast with the results of others, it was found that reducible cross-links of dental papilla (or pulp) were calcified tissue type and those of dental sac (stage I and III) were non-calcified tissue type. The excess acidic and basic amino acids were assumed to come from non-collagenous proteins. A dramatic change was demonstrated in the amounts of these amino acids at the three stages of dental papilla (or pulp) and dental sac.
