Abstract : 「まえがき」Eggenberger(1928)は, スイスの甲状腺センターAppenzellに於いて先天異常を見つけ, 唇裂, 口蓋裂をはじめとする多くの症例を報告し, これらの先天異常は甲状腺起原のものであろうと述べている. その後, Bloss(1937), Pasma(1948), Hodges et al(1952)とElphinstone(1953)などが母親の甲状腺機能低下症が, 子供の先天異常の原因であることを指摘している. Langmanとvan Faassen(1955)は母親ラットの甲状腺を一部除去後妊娠させると胎仔の限の奇形の増加と唇口蓋裂が発現することを報告しているが, 唇口蓋裂の発現率については述べていない. 甲状腺ホルモンと唇裂発現の関係を取り扱ったWoollamとMillen(1960)は, "strong A"lineマウスを用い, 妊娠11日目と12日目にthyroxin(0.1mg/mouse/day)を投与したが, 18日胎仔では対照群と比較して唇裂の発現率が低下したと報告している.

Title : Incidence of Cleft Lip and Palate in CL/Fr Strain Mice by Maternal Partial Thyroidectomy : A Microscopic Observation

Abstract : Sixty-eight CL/Fr females were thyroidectomized and 30 dams were pregnant from which 191 fetuses were harvested. One hundred thirty three of them were non-cleft, while the remaining 58 fetuses were affected with cleft lip and palate. Forty-eight CL/Fr female mice received MMI(0.3mg/10g)at intervals of 24 hours for 4 days before pregnancy and the same dose of it was readministered on the 13th day(Group A)or on the 12th day(Group B)after pregnancy. In Group A 11 fetuses were affected with cleft lip and palate in the harvested 57 fetuses(cleft incidence:19.3%), while in Group B 22 fetuses were affected with cleft lip and palate in the harvested 80 fetuses(cleft incidence:27.5%). For control 184 non-cleft and 47 cleft fetuses obtained from unoperated 30 CL/Fr strain dams were used. No fetus with cleft lip alone was found in any of the fetuses in the present study. Three fetuses with cleft lip alone was found in the partial-thyroidectomized group, and one fetus with cleft palate alone was found in the MMI administered group A. Simonart band was found in 3 fetuses from the dams receivcd MMI, one of which a cleft was restricted from lip to the hard palate, the soft palate being closed. The incidence of cleft formation in Group B of MMI administered dams was much higher than that of the control group and the aborted fetuses were few in this group B. These findings might be led to conclude that the elevation or fusion of the palatal shelves was prevented due to MMI administered on the 12th day after conception. This conclusion, however, would not be acceptable, since only one fetus was affected with cleft palate in this group. Rodent cavity or cavities surrounded with degenerated tissue in the lip and palate regions are often found in the cleft samples in any of the groups. Therefore partial-thyroidectomy or MMI given to the dams may not have induced directly cleft formation in the lip or palate, but could have made the dams more susceptible to viral infection, then the affected parts of the embryos underwent degeneration which resulted in post-fusion rupture in the lip and/or palate.
