Abstract : 「緒言」ラット顎下腺で産生される, 上皮成長因子(epidermal growth factor, EGF), 神経成長因子 (nerve growth factor, NGF) 等の生理活性物質の生成は, androgenによって支配されている. このことから, ラット顎下腺が, androgen依存の臓器であることは, 広く知られている. 従って, 顎下腺の生理的動態を知るメルクマールとして, 顎下腺におけるandrogenの作用機構を確立することは, 非常に意義深いと考えられる. androgen標的組織でのandorogen作用の指標となるものは, androgen receptorの存在と, その性質である. receptorが存在しても, androgenとの結合能がないか, androgen-receptor複合体が核内で作用しなければ, そのandrogenの標的組織細胞内での意義は無くなる.

Title : Androgen receptor in rat submandibular salivary gland - The role of cytosol for the salt resistant dihydrotestosterone binding site in nuclei -

Abstract : [Abstract] : 3H-Dihydrotestosterone (3H-DHT) binding substance in the nuclear fraction of rat submandibular salivary gland (SMG) was investigated. In exchange assay, 3H-DHT binding was stable for 20 hours at 0℃ and for 9 hours at 15℃. In competitive assay, relative ability of various steroids to compete with 3H-DHT binding was in the following order : DHT >= Methyltrienolone > Testosterone > Progesterone > Estradiol. The specific binding of 3H-DHT appeared in the residue after extraction from nuclei with 0.5M KCl buffer solution, whereas the specific binding was not shown in the extract. The Kd and NBS values were estimated by saturation and Scatchard analysis. The salt resistant DHT binding substance in the rat SMG nuclei were 6.64nM and 1.24pmol/mg DNA respectively. These salt resistant DHT binding sites in nuclear fractions of rat SMG were also observed in the binding experiment of nuclei and cytosol prelabelled with 3H-DHT for 30 minutes at 37℃. The 4S receptor was found in the prelabelled cytosol by 10~30% glycerol density gradient centrifugation. Prelabelling of cytosol with 3H-DHT caused the specific binding increase of 3H-DHT in the salt resistant particle of nuclei. The results show that the 4S receptor and DHT complex, in vitro, may play a role of stabilizing or enhancing the DHT binding in the salt resistant binding site of the rat SMG nuclei.
