Abstract : 「緒言」歯槽骨は歯周靱帯を介して歯を歯槽窩中に固定保持し, 歯に加わる外部からの機械的刺激の影響を受けて常に活発な改造を続けている組織である. したがって, 歯槽骨細胞の活性や外部刺激に対応した機能変化は, 歯科医学において重大な関心事の1つであり, 歯槽性の歯の移動を日常的におこなっている歯科矯正学の分野ではとくに関心が高い. 歯の移動に伴っても生ずる骨の改造は, 骨の吸収と形成がそのバランスを保ちながら進行し, 新しい内部構造を作り上げていく現象である. この過程の調節は骨を構成している細胞群の働きによるものであるが, 骨改造の場で直接関与しているのは細胞の合成する蛋白質である. 骨組織はその乾燥重量の20~30%の蛋白質を含む. この蛋白質が骨組織に力学的なねばり強さを与える働きや骨の吸収, 形成を調節する働きをしている. さらに, 一般的には, 骨有機質の約90%はコラーゲンと考えられているが, 石灰化度の低い類骨組織(osteoid)では非コラーゲン成分の占める割合が高く, 全有機質量の31%を占めている.

Title : Isolation and Characterization of Osteonectin from Rabbit Alveolar Bone

Abstract : [Abstract]: Osteonectin is a major glycoprotein in bovine and porcine bones and is found associated with mineral phase of bone. The experimental study was undertaken to clarify the physiologic natures of alveolar bone noncollagenous protein in rabbit, especially those of osteonectin. Osteonectin of rabbit alveolar bone was purified by chromatography of the formic acid precipitate of an EDTA extract. Sequential chromatographic steps consisted of gel filtration on Sepharose CL 6B in 4M guanidium HCl, ion-exchange chromatography on DEAE-Sephacel in 4M urea and chromatography on Sephacryl S200. The preparation consisted of pure form osteonctin containing high amount of aspartic acid and glutamic acid. The amino acid composition of rabbit alveolar bone osteonectin was very similar to that from bovine bone. Apparent molecular weight of alveolar bone osteonection was identical to that from bovine bone which had apparent molecular weight at Mr = 40000 on SDS-PAGE. Alveolar bone osteonectin did not demonstrate appreciable binding to denatured collagen, although it had indicated that the osteonectin relatively strongly bound to hydroxyapatite crystals. These studies indicated that in rabbit alveolar bone osteonectin is one of the major noncollagenous proteins and may have a role in mediating the bone formation processes.
