Abstract : 「緒言」Ga-67 citrate(以下Ga-67とする)は, 悪性腫瘍および急性炎症の画像診断に広く利用されている. しかし, Ga-67の腫瘍への集積と放射線や抗癌剤治療との関係の研究はあまりみられない. 1984年BekermanらはHodgkin's diseaseと肺癌(squamous cell, small cell)に化学療法(adriamycin, vincristine, cyclophosphamide, VP-16-213, 5-FU)を行った後に, Ga-67シンチグラムを行い, 骨への取り込みは増加したが肝, 筋肉, 腫瘍への取り込みは減少したと報告している. 1985年Iosilevskyらは, 繊維肉腫を移植したマウスに抗癌剤のcyclophosphamideを投与し, 腫瘍及び各組織におけるGa-67の集積の変動について報告している. しかし, in vitroにおける細胞レベルでの抗癌レベルでの抗癌剤の作用とGa-67の腫瘍細胞への取り込みに関する研究は, ほとんどみられない.

Title : The Effects of Antineoplastic Agent (Adriamycin) on Ga-67 Uptake in Cultured Tumor Cells.

Abstract : [Abstract]: This is a report on the uptake of Ga-67 into cultured tumor cell using mouse leukemia cell (L5178Y) in relation to antineoplastic agent, adriamycin. The results are as follows : Adriamycin exerts a statistically significant stimulation upon the incorporation of Ga-67 into mouse leukemia cells and those cells that were pretreated with adriamycin exhibited a proportional increase of Ga-67 uptake depending on the concentration and duration of time. These pretreated cells also showed a significant increase of Fe-59 into the cells which was not more than that of Ga-67. The alteration of cell cycle, determined by Flow cytometry, appears to be the major mechanism by which adriamycin carries out accumulation of G2 stage of cell cycle within 15hours after the treatment. However, treatment of the cell with actinomycin D has shown to decrease Ga-67 uptake without alteration of the cell cycle. Electron micrograph for adriamycin treated cells shows the enlargement of the cell size with a disappearance of microvilli from the surface of the cell. Therefore, the cytometrical determination of the cell cycle seems to be increasingly important with regards of Ga-67 uptake.
