Abstract : | 「緒言」ヒト赤血球膜の細胞骨格, 即ち, 裏打ち構造の分子構築については, タンパク質化学的にも超微形態学的にもかなりのことが明らかにされており, その機能についても, 形態変化, 変形能, 膜の安定化や, 膜内粒子の分布調節等に関して多くの議論がなされている. 近年, われわれは, ヒト赤血球膜のCa2+およびMgATPに依存した形態変化が, 裏打ち構造体(triton shell)でも同様に生ずることを見いだした. しかしながら, この現象は低イオン強度下においてのみ認められ, 生理的イオン強度下ではTriton shellは直ちに収縮し, Ca2+やMgATPに対する反応は観察し得なかった. 一方, ヘモグロビンは赤血球膜のATPによる形態変化の速度や, 赤血球膜の容積, また裏打ち構造の主要構成タンパクであるスペクトリンの二重体-四量体平衡に影響を与えると報告されている. |

Abstract : | [Abstract]: Triton shells shrink reversibly, depending on ionic strength, and their size is fairly small in 0.154 M KCl. However, when a hemolysate supernatant was added to a shell suspension in an isotonic medium, the shells swelled. The effect of heating on the hemolysate supernatant showed that there were two components which acted on the shells ; one was heat-labile and the other was heatstable. The heat-labile factor was identified as hemoglobin. The heat-stable factor was 1) stable after heating at 100℃ for 240 min, 2) not affected by HClO4, 3) precipitated with ethanol, 4) not soluble in chloroform-methanol and 5) inactivated by NaIO4 treatment. These two factors may regulate functions of the cytoskeleton in intact erythrocytes. |
