Abstract : 「緒言」halothane麻酔中, 血圧低下, 徐脈および末梢血管の拡張がみられることはよく知られている. これらはhalothane濃度と密接な関係があり, 低濃度においてはその変化は少ない. 末梢血管の拡張は, halothane投与直後から始まり, 皮膚表在性静脈の拡張としてとくに観察される. この状態は, 出血や手術侵襲の有無と無関係に麻酔の全経過を通じてみられる. halothane麻酔による徐脈は迷走神経のover activityによるものであると理解されているが, 末梢血管の拡張の原因に関しては, 血管平滑筋に対する直接作用, 血管運動中枢の抑制, 節伝達の抑制, 副腎髄質からのcatecholamineの放出抑制など多種多様の報告がありいまだに定説がない. そこで, 本研究では, 摘出イヌ腸間膜動脈および静脈を用いて以下の観点からhalothaneの効果を薬理学的に検討した.

Title : Effect of Halothane on Isolated Canine Mesenteric Artery and Vein - On the Hypotensive Effect of Halothane During General Anesthesia -

Abstract : [Abstract]: This study was designed to investigate the mechanism of the effect of halothane on isolated canine mesenteric artery and vein. The experiments were performed on isolated mesenteric arteries and veins from mongrel dogs (10-25kg) anesthetized with sodium pentobarbital. After the perivascular tissue was dissected away, the artery and vein were cut into rings which were suspended for isometric tension measurements. In the concentrations used (1.0, 2.0, 4.0%), halothane did not alter the basal tension of the vessel rings. Halothane (1.0, 2.0, 4.0%) had no effect on the norepinephrine (5×10-9-5×10-6 M) - induced contraction of the artery and vein. However, maximum contraction induced by transmural electrical stimulation were lessend by halothane (1.0, 2.0, 4.0%) in the artery and vein. Acetylcholine (10-9-10-5 M) produced endothelium-dependent relaxation of artery. This effect of acetylcholine was not influenced by halothane at the concentration tested. Acetylcholine produced concentration-related contraction of intact or endothelium removed veins. Observed contracting effect of acetylcholine on the vein was significantly inhibited by halothane (2.0, 4.0%). These results suggest that, 1) halothane has not α-adrenoceptor blocking properties ; 2) halothane inhibits norepinephrine release from sympathetic nerve ending ; 3) halothane may cause peripheral venodilation via endothelium-independent inhibition of the contracting effect of acetylcholine on the vein.
