Abstract : 「緒言」成熟マウス顎下腺は, 種々の生物学的活性物質を含有していることが知られており, これらの物質は一般に顆粒管細胞内に局在することが確かめられている. この生物学的活性物質であるEpidermal growth factor(EGF)とNerve growth factor(NGF)は, 顎下腺内に多量に存在し, その生理的役割から興味深い研究対象とされている. EGFとNGFの含有量は, 成熟マウスにおいて量的な雌雄差が認められ, 雄は著しく多く雌では少ないことが報告されている. またこれらの含有量は, Lacassagneが始めて報告したマウス顎下腺の顆粒管における性的差異と関連した変動を生ずることが明らかにされており, 両factorの合成, 貯蔵, 分泌およびホルモン調節等について, これまでに多くの研究がなされている. 生下時のマウス顎下腺は, 未熟なsecretory unitであるTerminal tubuleと未発達な導管系によって小葉が構成され, EGF, NGFは生後初期においては証明されず, 腺組織の分化発達に伴って次第に増加することが確かめられている.

Title : Immunohistochemical Studies of Epidermal Growth Factor and Nerve Growth Factor in Submandibular Glands of Mice During Period of Pregnacy and Lactation

Abstract : [Abstract]: The morphological changes of the submandibular gland (SMG) during the period of pregnancy and lactation were studied in the ddY mice. SMG was followed from 7 days of pregnancy through 21 days of lactation (just prior to weaning). Sections of the glands were stained with hematoxyline-eosin and immunocytochemically the avidin-biotin peroxidase complex method, for the localization of epidermal growth factor (EGF) and nerve growth factor (NGF). During the pregnacy and the lactation the granular convoluted tubules (GCT) of the ductal portion in SMG were larger and more numerous than that was found in control (virgin and non-pregnant) mice. These GCT cells exhibited increased numbers of cytoplasmic secretory granules. The immunoreactivity of EGF and NGF was detected in the GCT cells. In the glands of controls, there was a cell to cell variation in the intensity of immunostaining. In pregnant and lactating mice, most of the GCT cells were stained strongly with EGF and NGF immunostaining. According to the morphometric analysis, in the glands of pregnant and lactating mice the proportion of immunoreactive cells was significantly higher than that of controls. The ductal element occupied area in SMG increased in pregnant mice to 5.7-fold and in lactating mice up to 4-fold. Moreover, the individual GCT cells revealed an increase in the mean cell size relative to these cells in the gland of controls.
