Abstract : 「緒言」歯周病の病因に関する研究において, 異なる病型の歯周病には, それぞれ特異的な細菌あるいは細菌群が関与していること, あるいは歯周病に認められる組織破壊は, 細菌由来の抗原やマイトゲンに対する特異的あるいは非特異的な免疫応答によって誘起される可能性のあることなどが明らかにされてきている. Newman(1976), Slots(1979)およびTanner(1979)らは, さまざまな病型の歯周病患者の歯周ポケット内細菌叢を健全者のそれと比較して, グラム陰性嫌気性桿菌の占める割合が著しく高いことを指摘するとともに, その病因的意義について示唆している. すなわち, 若年性歯周炎(Juvenile periodontitis)ではActinobacillus actinomycetemcomitansとCapnocytophaga species, 急速進行性歯周炎(Rapidly progressive periodontitis)では, 主としてBacteroides gingivalis, Fusobacterium nucleatumおよび口腔スピロヘータ, さらに成人性歯周炎(Adult periodontitis)では, B.gingivalisあるいはBacteroides intermedius, Eikenella corrodens, そして口腔スピロヘータなどが高率に検出されることが明らかにされている.

Title : Chemical Structure and Immunobiological Activities of Peptidoglycan Isolated from Bacteroides gingivalis

Abstract : Abstract : The chemical structure and immunobiological activities were investigated on the cell wall peptidoglycans isolated from Bacteroides gingivalis. Peptidoglycans were isolated from B.gingivalis strains 381 and 1021 by boiling in 4% sodium dodecyl sulfate and by digestion with pronase and trypsin. Analysis of amino acids and amino sugars of the peptidogylcans revealed that muramic acid, glucosamine, D-glutamic acid, D-alanine, L-alanine and meso-2, 6-diaminopimelic acid (A2pm) were the principal components. Additionally, serine and glycine were found as minor components. Dinitrophenylation method revealed that about half of A2pm residue had a free amino group, and analysis by hydrazinolysis showed that a small part of alanine, A2pm, and glycine located at the C-terminal. The above results indicate that one of the amino groups of A2pm residue at one strand of the stem peptide subunit crosslinked to the carboxyl group of the L-amino acids as like as serine and glycine. It was thus revealed that the peptidoglycan of B.gingivalis belonged to the A3γ type of the classification by Schleifer and Kandler. Peptidoglycans isolated from B.gingivalis strain 381 and 1021 were found to be definitely adjuvantactive in induction of delayed type hypersensitivity against ovalbumin when administered to guinea pigs as water-in-oil emulsion and stimulation of increase sesum antibody levels was found in both peptidoglycans. Regarding mitogenicity on splenocytes of BALB / c and BALB / c nu / nu mice, peptidodglycans from two strains of B,gingivalis were markedly enhanced the uptake〔3H〕thymidine in dose of 10μg/105 cells, however thymocytes were not reactive. Stimulation effects on peritoneal macrophages from a guinea pig to incorporation of 14C-glucosamine were exhibited on addition of 10μg of both peptidoglycans. These findings indicate that peptidoglycan of B.gingivalis might eventually be responsible for destruction of periodontal tissue by host mediated activities.
