Abstract : 「序論」 骨組織は, 通常骨形成過程と骨吸収過程が平衡を保つことにより維持されている. その骨の生理や骨に対するホルモンの影響についての研究には, in vitroの方法が用いられていることが多い. 最初にin vitroで骨吸収を証明したのは18~19日の胎仔ラットの長幹骨を用いたもので, ホルモンや無機イオンと骨吸収との関係の解明に貢献した. しかし, 新たにつくられる骨組織については注目されなかった. また研究材料として, 胎仔マウス頭蓋冠が用いられ, 骨吸収に関与する多くの物質の生理学的作用の解明に役立っているが, 骨吸収と比べると, 骨形成に関する情報は少なく, 1980年代に入ってようやく多くの生理, 生化学的情報が供給されるようになった. in vitroで骨形成に注目した最初のこころみは, 1960年初めに5日鶏胚の大腿骨原基を約1ヵ月間培養し, 生理的骨形成が起こったことを報告したFellらの仕事によるものである.

Title : Ultrastructural Changes in Mononuclear Cells after The Treatment of PTH, PGE2, or Vit D3 in vitro

Abstract : Abstract : Utilizing conventional light, electron microscopy, histochemical and enzyme-antibody analysis, this study was carried out to investigate the morphological changes of osteoblasts and macrophages after cultivation for ten days under the addition of parathyroid hormone (PTH), prostaglandin E2 (PGE2) or Vitamine D3 (Vit D3) respectively, using the parietal bone fragments of fetal mouse calvaria (ICR strain). After the individual addition of PTH, PGE2 or Vit D3, osteoblasts surrounding the calcified bone was grouped into two types : The first appeared as a compressed profile and contained a few organelles. The second had a well-developed organelles, abundant dense bodies, expanded r-ER, and an increased mumber of mitochondria. Regarding the macrophages, they showed no particular morphological changes except an increased number of dense bodies, mitochondria and developed microvilli compared to the control group. With the addition of PTH, a large increase in the number of active macrophages were observed. With the addition of PGE2 no significant change compared to the control group was shown. Lastly, the addition of Vit D3 manifested an increase of the macrophages similar to the PTH group. The Vit D3 group also exhibited a fusion of mononuclear cells with cach other, resulting in the formation of multinuclear cells. This finding was not exhibited in the PTH and PGE2 groups. The presence of ACPase activity in these dense bodies suggests that they are associated with the lysosome. By the mixture of glutaraldehyde and tannic acid in the prefixative, multilamellar structures (MLS) with a regular periodicity were found both in the cytoplasm and on the cell membrane of the osteoblasts throughout the experiments. Further investigations are required to clarify the role of the MLS in the calcification process.
