Abstract : 「緒言」 小児歯科における成長発育, すなわち無歯期から始まり乳歯の萌出, 乳歯列完成そして吸収・脱落などの生理的変化を経て, 混合歯列期から永久歯列期へ移行する. その中でも, 特に歯周組織の成長過程を解明することは重要なテーマの一つである. 歯周組織, 特に歯根膜組織は歯小嚢から由来している. 斎藤らはヒト歯根膜組織片からexplant法により, ヒト歯根膜線維芽細胞(Human Periodontal Ligament Fibroblast: HPLF)の培養が可能となった. その結果, HPLFはアルカリフォスファターゼ活性(ALP活性)を示すことから, 皮膚及び歯肉の線維芽細胞と異なることが明らかとなり, osteoblastic fibroblastと提唱した. また, SomermanやCarnesはヒト歯根膜中にALP活性の高い線維芽細胞が存在することを報告している.

Title : The Cytobiochemical Study of Periodontal Ligaments of Human Deciduous Teeth - Cell Growth Regulatory Factors in Conditioned Medium -

Abstract : [Abstract] The processes of tooth development including the resorption of deciduous tooth and exchange of permanent tooth are regulated by various growth factors but their mechanism remained still uncertain. Human periodontal ligament-fibroblats of deciduous tooth (HPLF-Y) secret factors which stimulate the cell proliferation and regulate the alkaline phosphatase (ALP) activity of HPLF-Y and human periodontal ligament fibroblasts (HPLF). HPLF-Y and HPLF were obtained by the explantation method described by S.Saito et al (Adv Dent Res, 1988). HPLF-Y were inoculated. And cultured with D-MEM supplemented with 5% FCS, 50μg/ml ascorbic acid and antibiotics until confluent period and then, the conditioned medium (CM) of HPLF-Y cultured with FCS-free medium was collected. CM of HPLF-Y was fractionated with PO-60K of gel chromatography eluted with PBS (-) containing 0.1mM PMSF. After the total CM and fractionated CM of HPLF-Y were added to the HPLF-Y and HPLF, ALP activity and DNA content were determined by the methods using p-nitrophenylphosphate and diaminobenzoic acid, respectively. And also, the effect of CM with or without FCS on the cell proliferation of quiescent HPLF was performed by the incorporation of 3H-Thymidine. Though the ALP activity of HPLF-Y and HPLF was slightly decreased, the growth of HPIF-Y and HPLF was increased by the total CM of HPLF-Y. However, the fractionated CM of HPLF-Y didn't much stimulate the cell growth of HPLF, and increased the activity of ALP of HPLF. The CM of HPLF-Y stimulated the proliferation of quiescent cells with FCS. Therefore, the growth regulatory substances which were secreted by HPLF-Y, may modulate the cell growth and differentiation of HPLF-Y and HPLF.
