Abstract : 「緒言」 歯根膜は強靱な非石灰化の結合組織で, 歯槽骨とセメント質の石灰化組織の間に位置し, 間欠的な咬合力を受けている. 歯根膜はMelcher AHとTen Cate ARが報告している様に, 歯槽骨とセメント質と同じく, 歯小嚢由来である. 従って, Roberts WE and Chase DCとMcCulloch CAG and Melcher AHは歯根膜組織に線維芽細胞のprogenitorばかりでなく, 骨芽細胞とセメント芽細胞のprogenitorsの存在を示唆する報告をしている. 一方, 近年, Saito S et al, Kawase T at alと川瀬らにより, 歯根膜由来の線維芽細胞(HPLF)の培養系が確立され, この細胞の特徴からosteoblastic fibroblastと提唱した. 青木は活性型ビタミンD3である1,25(OH)2 D3によってHPLFのalkaline phosphatase(ALPase)が上昇し, 石渡はHPLFにALPaseと異なる phosphotyrosine phosphatase(PTPase)の存在を示し, HPLFの接着・伸展において細胞外基質成分のリン酸化と共にPTPaseが関与していることを報告している.

Title : Cell Attachment and Spreading Factors of Human Periodontal Ligament Fibroblasts (HPLF)

Abstract : [Abstract] Previous studies have suggested that human periodontal ligament-derived fibroblasts (HPLF) have osteoblastic characters related to alkaline phosphatase (ALPase), 1,25 (OH)2 D3 responsiveness and bone proteins synthesis. The purpose of this study is to clarify the property of cell attachment and spreading factors (CASFs) produced by HPLF. HPLF obtained by the explant method of Kawase T and Saito S were maintained with DMEM supplemented with 5% FCS and then cultured with FCS-free MCDB 107 medium for 24 hrs. The conditioned medium (HPLF-CM) was collected, condensed by the method of ultrafiltration (YM-5 membrane) and fractionated by the PO-60K column eluted with PBS (-) containing 0.1 mM PMSF. The cell attachment and spreading assay was performed by the inoculation of HPLF on the hydrophobic wells coated with total HPLF-CM, fractionated HPLF-CM, fibronectin (FN) and vitronectin (VN). Also, Gly-Arg-Gly-Asp-Ser (GRGDS) peptide was used for the inhibition assay of integrin receptor which has a binding site for the RGD signal CASFs. The DNA content of cells and ALPase activity were determined by DABA and p-NPP methods, respectively. HPLF-CM did not stimulate only the proliferation, but also increased the ALPase activity of HPLF in the same manner of bovine periodontal ligament (BPL) extract and TGF-β. The molecular weights of CASFs in HPLF-CM could be estimated approximately 50KDa and 10KDa. The 10KDa CASF, which can bind the heparin, inhibited the cell attachment and spreading of HPLF by GRGDS peptide, but the 50KDa CASF was not. Therefore, HPLF-CM could contain the cell growth and differentiation factors. And, it was suggested that HPLF-CM possessed a potent cell attachment and spreading activity compared with 50% FCS, FN and VN.
