Abstract : 「緒言」感染根管内から高頻度に分離される偏性嫌気性菌は, 根尖歯周組織に対して種々の病原性を有し, 根尖性歯周炎の最も重要な病原因子と考えられている. 現在, これら偏性嫌気性菌の菌体成分であるLPS, ペプチドグリカンおよび可溶性タンパク, さらには, 細菌代謝産物である各種タンパク分解酵素や産生毒素などは, 根尖歯周組織に対して直接的に組織傷害を惹起させると同時に生体における免疫応答を引き起こし, 間接的に組織を傷害する可能性が示唆されている. 本研究でとりあげたVeillonella parvula(以後V. parvula)は口腔内に常在する偏性嫌気性のグラム陰性球菌であり根尖病変を有する根管内から高率に分離される菌である. 感染根管内からの本菌の分離頻度は極めて高く, 現在までに堀場らは, 30歯中21歯(77.8%)山本らおよび広畑は, 13歯中3歯(23.0%), 豊嶋らは, 14歯中3歯(21.4%), 谷らは, 40歯中6歯(15.0%)にそれぞれV. parvulaを検出している.

Title : The Effects of Products from Human Peripheral Monocytes Stimulated with Cellular Components of Veillonella parvula to Fibroblasts from Periodontal Ligament, Pulp and Gingiva.

Abstract : Abstract: It has been suggested that gram negative anaerobic bacteria in infected root canals and/or periodontal pockets are important agents in the destruction of periodontal tissues. This study was conducted to examine the effects of products from the peripheral monocyte stimulated with cellular components of Veillonella parvula in infected root canals on periodontal tissues. In this study, sonic extracts prepared from V. parvula, predominantly isolated from infected root canals, were tested for their ability to produce prostaglandin E2 (PGE3) and collagenase from human peripheral monocytes. Furthermore, the effects of supernatant of culture of peripheral monocyte stimulated with sonic extract of V. parvula on the PGE2 and collagenase production and alkaline phosphatase (ALP) activity of fibroblasts from human periodontal ligament (HPLF), pulp (HPF) and gingiva (Gin 1) were examined. The results obtained were as follows: 1. PGE2 production of the peripheral monocytes was increased when the cells were stimulated by sonic extracts of V. parvula, but collagenase production was not increased. 2. MCM stimulated with sonic extracts of V. parvula induced PGE2 production of periodontal tissue fibroblasts. The activity of PGE2 of HPLF was higher than that of other fibroblasts. 3. MCM stimulated with sonic extracts of V. parvula for 96 hours strongly induced collagenase production from HPLF and HPF, however, the Gin 1 cells did not produce collagenase. 4. ALP activity of HPLF and HPF did not increase by the addition of MCM stimulated with V. parvula. Furthermore, the Gin 1 cells did not entirely show the ALP activity.
