Abstract : 「緒言」口腔癌は組織由来が同じでも, その生物学的性状は多彩であり, 治療に対する反応態度も症例によりかなり隔たりがみられる. ことに放射線, 化学療法の適用に際しては, 副作用を回避し, 的確に治療効果を挙げるために, 治療効果を予測し, それを客観的に評価するための指標が求められる. なかでも細胞動態の把握は, 放射線や, 多くの化学療法がcell cycle依存性であることから, その選択にとって重要である. 細胞動態の解析には, 核酸量の細胞測光や, Ki-67, BrdU, PCNAなどの増殖関連因子の免疫組織学的検索がなされているが, 検索に際し, 特別な設備や検体処理法を要したり, 手技も煩雑なことなどから, 日常的に行うには制約がある. 近年, 通常の光顕用パラフィン組織切片上で検索可能なAg-NORs(argyrophilic nucleolar organizer regions)染色が注目されている.

Title : Study on the Cell Kinetics of Oral Squamous Cell Carcinoma - Ag - NORs and Proliferation in Culture Cell -

Abstract : Abstract : In order to obtain an objective index for the activity of proliferation of cancer cells in oral squamous cell carcinoma, cultured cells(Ca9-22)were used to study the relationship between intranuclear Ag-NORs(argyrophilic nucleolar organizer regions)number, the cell growth curve, the content of the cellular RNA and the structure of the nucleoli. The changes in Ag-NORs number induced by anticancer agents(PEP, CDDP, 5-FU)were also studied. Results of the study follow. 1. In ordinary cultures with serum(control), the intranuclear Ag-NORs number increased during the cellular logarithmic growth phase and decreased during the stationary phase. In cultures without serum, the intranuclear Ag-NORs number decreased in the accompany of a decrease of the number of cells. 2. In the controls, the cellular RNA content increased slightly later than a increase of the Ag-NORs number initially and both reached a peak 4 days after culturing. The changes of the Ag-NORs number and the RNA content were nearly identical. 3. The Ag-NORs number was inversely correlated(r0=-0.760)with the roundness of the nucleoli, and positively correlated(r0=0.743)with the area of the nucleoli. 4. The anticancer agents caused an inhibition of cellular growth and a decrease of the Ag-NORs number which depended on the concentration of PEP and CDDP and upon the length of exposure to 5-FU. 5. When exposed to 5-FU, the nucleolar roundness gradually incresed as seen in the controls but the RNA content remained lower than that in the controls. When exposed to PEP and CDDP, the nucleolar roundness remained greater from the early stages, but the RNA content remained higher dispite lower Ag-NORs numbers. These results suggested that the intranuclear Ag-NORs number might be a useful index for the activity of cell proliferation in oral carcinoma and the therapeutic effect of anticancer agents.
