Abstract : 「緒言」日常の歯科臨床, 特に補綴分野において, 失われた咀嚼機能の回復により, 口腔のみならず全身的な健康の回復・増進をもたらすことはしばしば経験することである. 全身的な健康に深く関連する咬合や咀嚼のシステムを生物学的見地から明らかにすることは, 歯科領域の重要な研究課題の一つである. 咀嚼時に歯周組織は常に間欠的な機械的外力を受けている. 咀嚼によって起こる機械的外力により歯周組織を構成する細胞, すなわち歯根膜線維芽細胞および歯槽骨骨芽細胞がいかなる応答を示すかを生化学的に明らかにすることは重要な事と思われる. 斎藤とSaito S et alに機械的外力を, 1: 物理的力として張力, 圧縮と重力, 2: 圧力として大気圧, ガス分圧と水圧, 3: 熱と光, さらに4: 電界と磁界のカテゴリーに分類しており, これらに対する生体の反応は, 1: 超微構造的, 2: 生化学的, さらに3: 生物物理的検索によって明らかになるものと報告している.

Title : Cellular Responses of Human Periodontium Derived Cells by Mechanical Stress

Abstract : Abstract: The aim of this study was to explore the effect of mechanical stretching on the cellular responses of human periodontal ligament fibrolasts (HPLF) and alveolar bone cells (HABC) obtained by the explant method described by Kawase et al (Adv Dent Res 1988). The cells were maintained with D-MEM supplemented with 5% FCS, 50μg/ml ascorbic acid and antibiotics. At a confluent phase of cultured HPLF and HABC, the culture medium was replaced with a FCS-free MCDB 107 medium. Cells were exposed to cyclic stretching using a conputerized, pressureoperated instrument of Flexercell strain unit with 3 cycles/min of tension which was calculated at 6, 18 and 24% elongation of the cells compared with their original confluent dimesion. After cells were stretched for 24 hrs, the cell attachment and spreading factor (CASF) activity of the HPLF-CM and the HABC-CM were assayed by microscopic observation and the DNA content according to Kubo and Kawase (Kanagawa Shigaku, 1991). The activity of alkaline phosphatase (ALPase) was determined by p-nitrophenyl phosphate (p-NPP) as substrate. Cyclic stretching increased DNA content and enhanced ALPase activity in HABC. However, the ALPase activity of HPLF was dramatically stimulated by the magnitude of stretching in a dependent manner of elongation. The CASF activity of HPLF-CM and HABC-CM was increased approximately 2.3 and 1.9 fold at 18% elongation compared with the control cells, respectively. Therefore, the cell growth and ALPase activity of HPLF exhibited different responses compared with that of HABC. Additionally, it was suggested that mechanical stretching could modulate the CASFs synthesis of HPLF and HABC dose dependently. These findings indicate that the remodeling processes of periodontium, including cell proliferation and differentiation could be closely associated with mechanical stresses such as occlusion and mastication.
