Abstract　： 「論文内容の要旨」歯周疾患の治癒過程を細胞レベルで明らかにすることは, 歯周組織の再生を積極的に処置するときに重要な情報である. 斎藤らはヒト歯根膜由来線維芽細胞(HPLF)の培養系を確立し, HPLFの特徴を明らかにしてきた. そこで, 本研究においてHPLFの培養上清中の細胞接着・伸展因子(CASFs)を分離し, 精製することを行った. HPLFはSaito et al(Adv. Dent. Res. 1988)の方法のexplant法を用いて得られた. 5～8代継代細胞をconfluentまで培養し, FCS-freeのMCDB 107培地と交換し, 24時間の培養を行い, その培養上清をconditioned medium(HPLF-CM)とした. また, 10μg/ml cycloheximideを添加し, 5時間までのHPLF-CMを採取し, CASF活性を位相差顕微鏡で観察した.
