Abstract : 「緒言」健全な顎の発育ならびに口腔機能の育成を目的とする小児歯科領域において, 成長発育過程での歯および歯周組織の重要性を細胞生物学的に解明することは必要不可欠である. しかも, 歯周組織の一部を構成する歯根膜は, 歯根の形成のみならず永久歯の萌出にも関与している. また, 乳歯歯根膜は顎骨の成長や生理的歯根吸収の際にも重要な役割を果たしている. 間葉系歯周組織を主体とする歯胚の基質形成にはアスコルビン酸が必須であることはBerkovitz BKにより報告されている. L-Ascorbic Acid(AsA)は, proα chainのprolineとlysineにhydroxylaseが作用して水酸化反応が起こり, hydroxyprolineとhydroxylsineとなる過程においてのcofactorとしてcollagen合成過程に必須なものであると報告している.

Title : Effects of L-Ascorbic Acid, L-Ascorbic Acid-2-phosphate and Growth Factors on the Cellular Responses of Fibroblasts Derived from Human Deciduous and Permanent Periodontal Ligaments

Abstract : Abstract: It is generally believed that ascorbic acid plays a direct role in the processes of development of periodontal connective tissues. Nonetheless, many important aspects have not yet been touched on. This study was attempted to explore the regulatory potential of L-ascorbic acid (AsA) comparing L-ascorbic acid-2-phosphate (AsA-P) on the cell differentiation, and cell attachment and spreading of fibroblasts (HPLF-Y, and HPLF) derived from human deciduous and permanent periodontal ligaments. AsA-P which is stable in biological fluid has closely similar effects as AsA on the cell growth and alkaline phosphatase (ALPase) expression of HPLF-Y and HPLF. AsA and AsA-P were observed the essential factors on the synthesis and secretion of cell attachment and spreading factors (CASFs) of HPLF-Y and HPLF. The CASF activity of HPLF-Y exposed with transforming growth factor-β (TGF-β), interleukine-1β (IL-1β) and dexamethasone (Dex) coupled with AsA or AsA-P was significantly stimulated, In addition, the CASFs of HPLF were increased by TGF-β, insulin-like growth factor-I (IGF-I) and insulin-like growth factor-II (IGF-II) coupled with AsA or AsA-P. But, platelet derived growth factor (PDGF) of a competence factor decreased the CASF activity of HPLF-Y and HPLF. Therefore, AsA could not regulate only the cell growth and differentiation, but also the CASFs of HPLF-Y and HPLF. These findings demonstrated that AsA and TGF-β could be the essential regulators of CASFs.
