Abstract : 「緒言」 小児の歯周組織の成長過程は, 無歯期から始まり乳歯の萌出, 乳歯列完成そして吸収・脱落などの生理的変化を経て, 混合歯列期から永久歯列期へ移行する. 中でも, 乳歯の吸収・脱落, 永久歯の萌出などの生理的変化を解明することは重要なテーマの一つである. 歯周組織, 特に歯根膜組織は歯小嚢に由来している. 斎藤らはexplant法により, ヒト歯根膜組織片から歯根膜線維芽細胞(Human Periodontal Ligament Fibroblast: HPLF)の培養法を確立し, その特徴について報告している. その結果, HPLFは高いアルカリフォスファターゼ活性(ALP活性)を有することから, 皮膚及び歯肉の線維芽細胞と異なることが明らかとなり, osteoblastic fibroblastと提唱した. さらに, SomermanらやCarnesらはヒト歯根膜中にALP活性の高い線維芽細胞が存在することを報告しており, 斎藤らの主張しているHPLFの特徴を裏付ける結果となった.

Title : Proliferation and Differentiation Regulated by Growth Factors on Human Periodontal Ligament Fibroblasts Derived from Deciduous and Permanent Teeth

Abstract : Abstract: Growth factors are a class of natural biological mediators that regulate many activities of developmental processes including proliferation, differentiation, motility and matrix synthesis of the cells. The purpose of this study was to evaluate the effects of insulin-like growth factor-I (IGF-I), insulin-like growth factor-II (IGF-11), interleukin-1β (IL-1β) and platelet derived growth factor (PDGF) on cell growth and alkaline phosphatase (ALP) expression as a make of cell differentiation of human periodontal ligament fibroblasts derived from deciduous teeth (HPLF-Y) comparing the human periodontal ligament fibroblasts from permanent teeth (HPLF). (1) IGF-I: The growth of HPLF-Y incubated with IGF-I was increased in a dose dependent manner. HPLF was responded by the high dose (100ng/ml) of IGF-I. The ALP activity of HPLF-Y and HPLF terated with IGF-I were not stimulated, but increased by a combination of IGF-I and 1α, 25(OH)2 D3 (D)3. (2) IGF-II: IGF-II stimulted the cell growth of HPLF-Y and HPLF in a dose dependent manner. (3) IL-1β: The response of HPLF-Y was different from HPLF by IL-1β. IL-1β suppressed the expression of ALP activity of HPLF-Y and stimulated the cell growth of HPLF. (4) PDGF: The proliferation of HPLF-Y and HPLF were dramatically enhanced by PDGF, but not the cell differentiation. The contractions of cell layers of HPLF-Y and HPLF were morphologically observed after the confluent conditions. These findings demonstrate that the growth factors regulate the proliferation and differentiation of HPLF-Y and HPLF. During the development of periodontium of deciduous teeth, the growth factors could play the important roles in formation of teeth.
