Abstract : 「緒言」 破骨細胞は造血幹細胞に由来する血球系細胞であり, CFU-GMを経て単核の前破骨細胞となり骨芽細胞の形態変化に伴い露出した骨面に誘導され定着後, 多核化して骨を活発に吸収するようになる. この破骨細胞の多核化機構については3H-thymidineの投与実験などから単核細胞の融合によると考えられている. しかし, この単核の前破骨細胞が単球・マクロファージまでに分化した単核食細胞系の細胞であるのか, あるいは単核食細胞系に属するがもっと未熟な細胞であるのか, さらに幹細胞の付近ですでに枝分れして独自の分化をとげた細胞であるのかなどについてはいまだ意見の分かれるところである. 単球・マクロファージは運動性および活発な消化機能を持ち, 融合して多核化するなど破骨細胞と類似した特徴を有しているが, 破骨細胞はFCレセプター, C3レセプターあるいは非特異エステラーゼや内因性のペルオキシダーゼを欠如し, 骨吸収抑制ホルモンであるカルシトニン(CT)に対して感受性があることなど単球・マクロファージとは多くの相違点が示されている.

Title : Relationship between Differentiation and Expression of Surface Antigen in Osteoclasts

Abstract : Abstract : The clonal preadipose cell line, MC3T3-G2/PA6, can support the differentiation of osteoclast-like multinucleated cells in response to 1α,25-dihydroxyvitamin D3 [1α,25(OH)2D3] or prostaglandin E2, in culture with mouse spleen cell. In this study, I examined whether the mouse osteoclasts is formed by using this co-culture system express the antigen (Mac-1, Mac-2, F4/80, MOMA-2, BM8) known to be expressed in the cells of the monocyte-macrophage lineage. Mac-1 and Mac-2 antigen were detected on mononuclear preosteoclast and osteoclasts, but F4/80 and MOMA-2 and BM8 antigens were not detected on mononuclear preosteoclasts or osteoclasts. Furthermore, a significant number of Mac-2 positive-F4/80 negative cells were detected. A part of the Mac-2 positive-F4/80 negative cells were also tartrate-resistant acid phosphatase (TRACP ; a marker enzyme of osteoclasts) positive. On the other hand, the peritoneal resident cell (Mac-2 negative) population can not induce TRACP positive colony formation, but that the thioglycolate-elicited peritoneal exudate (Mac-2 positive) showed a high level of TRACP positive colony formation. Furthermore, the higher the percentage of mature macrophage in the peritoneal exudates the higher the percentage of TRACP positive colonies. Resorption lacunae were formed on ivory slices in which bone marrow cells or peritoneal exudates were co-cultured with MC3T3-G2/PA6 cells in the presence of 1α,25(OH)2D3. These findings suggest that expression of the Mac-2 antigen can play an important role in osteoclast differentiation.
