Abstract : 「諸言」小児の健全な咬合の育成ならびに口腔機能の獲得を目的とする小児歯科学領域において, 成長発育の過程での歯および歯周組織の形成と改造の現象を細胞生物学的に解明することは重要な課題である. これらを調節する因子としてはホルモン, 成長因子, ビタミン等, 数多く上げられるが, ビタミン群の中でもビタミンAに着目した. すなわち, ビタミンAはβ-カロチンのプロビタミンAとして生体に取り入れられ, 活性型のall-trans retinoic acid(at-RA)および9-cis retinoic acid(9-cis RA)に代謝される. ビタミンAの欠乏症として夜盲症, 生殖機能の異常, 皮膚および粘膜上皮の異常が報告されている. 一方Rothmanらは, 妊娠前後にビタミンAを投与することは奇形が高い確立で発生することを警告している. すなわち, ビタミンAは外胚葉性の組織ばかりか, 外胚葉性中胚葉を標的組織として作用している.

Title : Cytobiological Studies of Fibroblasts Derived from Human Deciduous Periodontal Ligament - Regulation of Proliferation and Differentiation by Retinoic Acids -

Abstract : [Abstract]: Retinoic acids (RA), natural derivatives of vitamin A, have a wide spectrum of biological roles in growth and differentiation of tissues. We demonstrated the effects of all-trans retinoic acid (at-RA) and 9-cis retinoic acid (9-cis RA) on cell growth, alkaline phosphatase (ALPase) activity and protein synthesis of fibroblasts derived from human deciduous and permanent periodontal ligaments (HPLF-Y and HPLF). Cells migrated from the explanted human periodontal ligaments were cultures in 5% FCS D-MEM without ascorbic acid and exposed with at-RA or 9-cis RA. At-RA and 9-cis RA inhibited the cell growth. The ALPase activity of HPLF-Y was markedly stimulated by at-RA and 9-cis RA at concentrations of 10 nM and 1 nM, respectively. The ALPase activity of HPLF was stimulated by at-RA and 9-cis RA at the concentration of 10 nM, while 1,25 (OH) 2 D3 (D3) altered cell growth and differentiation of HPLF-Y and HPLF exposed to at-RA or 9-cis RA for 8 days. In the growth phase, HPLF-Y did not alter the incorporation of 3H-thymidine (TdR) into cold TCA precipitates of cell layers, but the DNA synthesis of HPLF was inhibited by both at-RA and 9-cis RA. In the preconfluent and confluent phase, at-RA and 9-cis RA decreased the incorporation of 3H-TdR in cold TCA precipitates of HPLF-Y and HPLF. The total incorporation of 35S-methionine (35S-Met) was markedly decreased by the addition of at-RA, 9-cis RA and D3. Extracellular proteins labeled with 35S-Met were separated in fractions by IEC-DEAE column eluted with 0 to 0.5 M NaCl in 20 mM Tris-HCl, pH 8.0 to obtain 3 fractions which were non-interacted, 0.1 M NaCl and 0.2 M NaCl eluted fractions. Both at-RA and 9-cis RA decreased 0.1 M NaCl elution fraction, and also 0.2 M NaCl elution fraction containing osteonectin, while D3 and at-RA increased the non-interacting fractions. Therefore, at-RA and 9-cis RA may modulate the cell differentiation and proliferation of periodontal ligament cells.
