Abstract : [Abstract] In order to understand the effects of physical stress on bone metabolism in growing individuals, we designed a mouse model for running exercise. For 20 or 30 days, animals in the experimental group were given running exercise ; 240m a day. At these time points, the dissected femora were analyzed radiologically and chemically. In the experimental group, a higher ratio of X-ray absorption was observed in the experimental mice which ran for 20 or 30 days. Also, Ca contents in femora of these animals were significantly greater than that of control animals. The contact microradiography indicated that the trabeculae in the experimental femora were much thicker and denser than those of control femora. Increased synthesis of proteoglycans shown by the internal labelling of dissected proximal end of femur with 35S-sulfate was more apparent on day 10 in the experimental animals, although the levels of extracted uronic acid per mg bone remained the same in two groups. These results suggest that running stimulates bone formation while maintaining the quality of bone matrix.
