Abstract : 「緒言」 Porphyromonas gingivalis(P.gingivalis)は, 歯周炎患者の歯周ポケットから高率かつ高頻度に分離されることが報告されており, 成人性歯周炎の有力な原因菌として注目されている. また, 歯周炎患者の本菌に対する血清抗体価が有意に高い値を示すことや本菌の増加と歯槽骨破壊の進行との間に相関が認められることなどからも歯周炎に関与する病原細菌と考えられている. P.gingivalisの病原性に関しては, 本菌の産生する種々のタンパク分解酵素が直接的に組織破壊を引き起こす可能性の他, 内毒素による組織障害や炎症の誘発, 更に免疫系を刺激することによるアレルギー性病変の惹起などの可能性が示唆されている. 菌体表層に線維状構造物として存在する線毛も免疫細胞や歯周組織を刺激してサイトカインの産生を誘発する活性歯周組織細胞への付着性を介して重要な役割を果たしていると考えられている.

Title : Construction of a NotI Restriction Map and a Physical Map of Porphyromonas gingivalis Chromosomal DNA

Abstract : [Abstract] To establish the molecular basis of the virulence of Porphyromonas gingivalis ATCC 33277, a NotI restriction map of the chromosome was constructed by exploiting NotI-linking clones and DNA probes from various genes of P.gingivalis strains. The restriction endonuclease NotI was found to have digested P.gingivalis ATCC 33277 chromosomal DNA into 10 fragments ranging from 30 to 690 kilobases by pulsed field gel electrophoresis (PFGE). Total chromosomal DNA of P.gingivalis ATCC 33277 was estimated to constitute 2595 kilobases. Linking clones containing a NotI site were used as DNA probe for Southern hybridization to the fragments digested with the same enzyme. The probe was constructed to overlap two adjacent large DNA fragments. Thus it hybridizes to both of them and demonstrates that these two fragments must be adjacent in the chromosome. Since the initial cloning of the fimbrillin gene, other P.gingivalis genes encoding virulence factors have been cloned in E.coli. The direct assignment of the 6 virulence genes was identified to the map of the chromosome of ATCC 33277. The NotI fragment A hybridized with DNA probes for fimbrillin gene, 53-kDa outer membrane protein gene and hemagglutinin gene. Fragment B hybridized with DNA probe for Gry-Pro peptidase gene. The NotI fragment E hybridized with DNA probes for superoxide dismutase and 40-kDa outer membrane protein gene. This map facilitates genetic and molecular studies in P.gingivalis, and its construction serves as a model for further endeavors on larger genome. The NotI map of P.gingivalis ATCC 33277 and the approach for the direct physical mapping presented in this study will be useful in establishing the relationship of virulence-associated factors of this organism.
