Abstract : 「緒言」 破骨細胞は骨吸収の場に出現する多核巨細胞で, 血液幹細胞由来であることが明らかにされている. 骨改造現象に見られるような生理的骨吸収においても, また炎症や機械的刺激を原因とする病的骨吸収においても, 破骨細胞は単独で分化してその機能を発揮するのではなく, 骨芽細胞のような造骨系細胞やストローマ細胞の存在およびそれらの細胞との直接的な接触が必要であるといわれている. 歯周炎に伴っておこる歯槽骨吸収においても同様の機構が働き, 歯周組織局所において破骨細胞前駆細胞と分化支持能を有する細胞が接触して破骨細胞が誘導されると考えられる. 最近, 株化されたストローマ細胞を用いた実験系などから, 破骨細胞の分化や骨吸収のメカニズムさらに歯周病原細菌の骨吸収への影響などが明らかにされてきた. しかしこれまで使用されてきた破骨細胞分化支持能を有する株化細胞のほとんどが頭蓋冠もしくは骨髄由来であり, 歯周組織由来のものは少ない.

Title : Isolation and Characterization of Mouse Periodontal Ligament Cells Supporting Osteoclast Differentiation

Abstract : [Abstract] Osteoclast progenitors proliferate and differentiate into osteoclasts through cell to cell interaction with osteoblastic stromal cells. In this study, a mouse periodontal ligament cell, designated CP 1-27 that had the capacity to support osteoclast differentiation, was isolated and characterized. When mouse bone marrow cells were cultured with CP 1-27 in the presence of 1 α, 25-dihydroxyvitamin D3 [1 α, 25 (OH)2 D3] and dexamethasone, tartrate-resistant acid phosphatase (TRAP) positive mononucleated and multinucleated cells were formed. No TRAP positive cell appeared in the absence of 1 α, 25 (OH)2 D3 and dexamethasone. When mouse bone marrow cells and CP 1-27 cells were cocultured on an ivory slice in the presence of 1 α, 25 (OH)2 D3 and dexamethasone, resorption lacunae were formed. After confluence, they showed a high alkaline phosphatase (ALP) activity. However when CP 1-27 were cultured for 30 days, even with b-glycerophosphate and dexamethasone, no mineralized nodule was formed. The expression of typical osteoblastic bone matrix proteins namely osteocalcin, osteopontin and osteonectin were examined. The messenger RNAs of osteonectin and osteopontin were expressed in CP 1-27 but that of osteocalcin was not detected. The expression of vascular cell adhesion molecule-1 (VCAM-1) and intercellular adhesion molecule-1 (ICAM-1) were observed in CP 1-27. With the addition of anti VCAM-1 and anti ICAM-1 in the coculture, both the number of TRAP positive cells and the ratio of TRAP positive multinucleated cells were significantly decreased. These results suggest that a part of the periodontal ligament cells, their character is different from the osteoblast, is capable of supporting osteoclast formation and differentiation.
