Abstract : 「緒言」 骨吸収を伴う辺縁性歯周炎や根尖性歯周炎は口腔常在菌の感染により発症し, 病変の進展には感染に伴う起炎性物質の産生と同時に宿主の免疫応答が関与している. 多くの研究者により炎症局所の細胞間伝達物質として種々のサイトカインが重要な役割を担っていることが明らかにされている. サイトカインは通常生体を防御するために働くが, 感染によるLPSなどの細菌成分の刺激により歯周組織に存在する種々の細胞より産生され複雑なネットワークによって相互に干渉し, 組織破壊や歯槽骨吸収に関与している. IL-1, TNF, IL-6は免疫応答および骨吸収に密接に関与し, 歯槽骨吸収を伴う歯周炎の発症, 進展に関与している. また, ヒトやラットの歯周炎局所においてこれらのサイトカイン産生の局在が報告されている. 歯周炎は辺縁歯肉の炎症に始まり上皮付着が破壊され, 続いて歯根膜腔を介して炎症が波及し, 歯周ポケットが形成され歯槽骨の破壊が起こることから, 歯根膜組織を構成する細胞群やそれらの細胞より産生されるサイトカインが組織破壊に密接に関与していると考えられる.

Title : Effects of Inflammatory Cytokines on Osteoclasts Differentiation Supported by Stromal Cells Derived from Mouse Periodontal Ligament

Abstract : [Abstract] Inflammatory cytokines IL-1 α, IL-6 and TNFα, are involved in a bone resorption in periodontal disease and those are released from periodontal tissue. In this study, we determined the effect of these cytokines on TRAP positive multinucleated cell formation in coculture of mouse bone marrow cells and a mouse periodontal ligament cell (CP 1-27) that had a capacity to support osteoclast differentiation. The addition of these cytokines, many TRAP positive multinucleated cells were formed and the ratio of TRAP positive multinucleated cells were increased depending on the cytokines concentration. These results suggest that IL-1 α, IL-6 and TNFα play important roles in alveolar bone destruction caused by periodontal disease.
