Abstract : 「緒言」 従来, 脳への多様な刺激入力に対する活性化部位を同定するための研究手法として, 微小電極法などの電気生理学的な手法を始め, [14C]2-deoxyglucose法やFos様免疫染色法などによる解明が行われてきた. 最近では, 脳の核医学診断においてfMRI (functional magnetic resonance imaging), SPECT (single photon emmision computed tomography), PET (positron emmision tomography)などを使用することで直接生体での脳機能を観察することも可能となり, 解像度や感度の面でも急速に向上している. 一方, In situ hybridization組織化学法(ISH法)により, cRNAプローブなどを使うことで標的遺伝子のmRNA発現を検出することが可能である. つまり, 生体において, ある行動や刺激に対する脳内応答を遺伝子発現を介して探ることができる.

Title : Detection of c-fos mRNA Expression in Response to Stimulation by In Situ Hybridization (ISH) Method -Feeding Behavior and the Expression of c-fos mRNA in Rat Brain-

Abstract : [Abstract] In order to understand how feeding behavior stimulates the brain, the expression of c-fos mRNA was analyzed in rat brain in a temporal sequence and spacial distribution. The messages were detected by in situ hybridization (ISH) with 35S-labeled c-fos cRNA probe and visualized using BAS 3000 imaging analyzer (Fuji Film Co.). Induction of c-fos mRNA expression was demonstrated bilaterally 60 min after feeding in the caudal subnucleus of the spinal trigeminal nucleus (NTST), nucleus of the solitary tract (SOL), the ventromedial hypothalamic nucleus (VMH), the paraventricular hypothalamic nucleus (PVH), the amygdaloid nuclei (AMG), and the cerebellar cortex. The temporal and spacial expressions of c-fos mRNA were consistent with the previously published results in the feeding related stimulation. Furthermore, the increase in c-fos mRNA expression was also demonstrated bilaterally in the hippocumpus (HPC) where memories are processed. It is concluded that ISH method can detect very early responses in stimulated cells with c-fos mRNA which is one of the typical immediate early genes ; thus, ISH method proves extremely useful to visualize the transduction during the stimulation of brain.
