Abstract : 法医実務上, 身元不明死体の個人識別は常に重要課題として検討され, その手段には多くの報告がみられる. 特に, LandsteinerによるABO式血液型は, 法医学上, 個人識別の他, 異同識別, 親子鑑定および犯罪捜査などに利用され, きわめて重要で応用範囲が広く1)司法解剖時に行われる基本的な検査でもある. ABO式血液型判定は, 新鮮血のみでなく微量かつ腐敗, 陳旧化している分析試料に対しても用いられ, 吸収試験法, 解離試験, 混合凝集反応が代表的である. 特に, 解離試験は, 血液型抗原性の少ない組織からの検出が可能であることから硬組織の血液型判定に有効とされており2), 骨, 毛髪, 爪, 歯等はそれらが検査試料となる高度腐乱死体, 白骨死体等のABO式血液型判定に広く用いられている. 近年, 法医学領域におけるDNA分析の発展はめざましく, 特に, 目的とするDNA断片を短時間で正確に増幅することができるpolymerase chain reaction(PCR)法の応用は, 微量かつ腐敗, 陳旧化している分析試料からでもDNA分析が可能になった.

Abstract : In forensic practice, ABO-blood group genotype determination is the most fundamental work involved in personal identification. However, in forensic autopsy, postmortem changes, particularly putrefaction, make examination of the ABO system in tissues difficult, and interpretation of the results is even more troublesome. ABO-blood group activities in such samples have been detected using the absorption-elution test, which is ultimately based on the ability of indicator cells to agglutinate. This agglutination reaction requires a relatively large amount of antigen, and if the antigen is present in very small amounts, considerable skill is needed to determine the ABO-blood group. Therefore, the absorption-elution test does not always provide sufficient data for a final evaluation. Polymerase chain reaction (PCR) analysis offers the advantages of greater speed and sensitivity. ABO genotype can be analyzed by PCR of genomic DNAs and digestion of the PCR products with restriction enzymes at specific nucleotide cleavage points. We collected samples of teeth (5), and also muscle tissue that had been kept frozen at -20℃ (13 samples), fixed in formalin (8), or embedded in paraffin (1) for 5 years since the time of judicial autopsy, and examined their ABO genotypes utilizing PCR and restriction enzymes. As a result, it was possible to determine the ABO genotypes of all samples. PCR analysis allows clear determination of ABO genotype by specific bands. Thus, in forensic practice, PCR analysis can be used instead of the absorption-elusion test for the determination of ABO genotype.
