Abstract : 歯原性角化嚢胞は比較的高い確率で再発する傾向があり, 他の歯原性嚢胞と比較して顎骨内への進展など臨床的に活発な性格を示す. 嚢胞の臨床的性格を決定する因子として, 嚢胞壁上皮の増殖活性とアポトーシスが考えられている. 我々は歯原性角化嚢胞の上皮下結合組織中の線維芽細胞がHGFを産生し, 嚢胞壁上皮に発現したレセプターであるc-Metと反応することで, 上皮細胞の増殖活性が高まることを報告している1). 一方, 歯原性角化嚢胞の基底層から傍基底層にはBc1-2が発現し2, 3), 中間層からBc1-xL, Baxが発現することで, 嚢胞壁上皮のアポトーシス活性が調節されている可能性を報告している4). HGF/c-MetとBc1-2ファミリーとの間で相関があることが最近の研究で報告されている. 乳癌, 腎尿細管上皮においてHGF/c-MetがBc1-xLの発現を上昇させることでアポトーシスを抑制する現象がin vtroで確認され5), その後バーキットリンパ腫でも同様の現象が観察されている6).

Title : Expression of Apoptosis-related Factors in Odontogenic Keratocyst

Abstract : Proliferative activity and apoptosis of the lining epithelium are important determinants of the clinical characteristics of odontogenic keratocysts. We previously showed that HGF increased the proliferative activity of epithelial cells. We also demonstrated that the apoptosis-related factors bcl-2, bcl-x, and bax are expressed in the lining epithelium of cysts. A close correlation between the expression of HGF/c-Met and that of the bcl-2 family of proteins has been defined. To test the hypothesis that HGF/c-Met participates in expression of the bcl-2 family of proteins, we immunohistochemically studied the expression of HGF, c-Met, bcl-2, bcl-x, bax, and bag-1. HGF was found to be expressed by fibroblasts in connective tissue of odontogenic keratocysts. C-Met, bcl-2, bcl-x, bax, and bag-1 were expressed in the lining epithelium of cysts. The HGF receptor c-Met was expressed from the parabasal layer to near the superficial layer of the lining epithelium. In contrast, bcl-2 was expressed mainly in the basal and parabasal layers, and bcl-x was expressed mainly from the intermediate layer to the superficial layer. Sites expressing bax were similar to those expressing bcl-x, and bag-1 was expressed in the intermediate layer of the lining epithelium. Correlation analysis showed that the expression rates and staining properties of c-Met correlated with those of bcl-xL from the intermediate layer to near the superficial layer of the lining epithelium. These results suggest that the expression of bcl-xL is regulated by HGF/c-Met interactions.
