Abstract : 内分泌攪乱化学物質(いわゆる環境ホルモン)は, 内分泌機能を変化させ, 野生生物やヒトの健康に有害な影響を及ぼす外因性の化学物質である. 女性ホルモンの一種である17βエストラジオール(E2)はエストロゲンレセプター(ER)と結合しホルモン作用を起こすが, 環境ホルモンもこのERを標的の一つとし, 正常なホルモン作用に悪影響を及ぼす. 環境ホルモンのヒトに対する影響については, 精子数の減少, 乳ガン, 子宮内膜炎の増加と不妊などが指摘されている. 歯科領域では, Oleaらによって予防填塞材(フィッシャーシーラント)から唾液中に, 環境ホルモンの一種であるビスフェノールA(BPA)が高濃度に溶出されたと報告され注目されるようになった. しかしBPAを出発原料とするBis-GMAはフィッシャーシーラントやコンポジットレジンのベースモノマーとして広く用いられているが, これらからのBPAの溶出については, 微量検出の報告7)はあるが, 現在一般的にHPLCでは, 検出限界以下であると報告されている.

Title : Relationship between the Molecular Structures and Estrogenic Activity of Bisphenol A and its Chemical Analogs

Abstract : Many dental materials such as denture resins and orthodontic plastic brackets contain polycarbonates. Some of these polycarbonates, including bisphenol A and related compounds, are reported to affect on the endocrine system. We report here a correlation between the molecular structure of bisphenol A andthe estrogenic activity of these compounds with respect to the human breast cancer cell line MCF-7. We measured the amount of bisphenol A and bisphenol S contained in two dental resins (polycarbonate and polysulfone) by dissolving them with PBS (-) and analyzing the solution by high-performance liquid chromatography (HPLC). We examined the estrogenic activity of bisphenol A, its chemical analogs, and various phthalates by measuring cells growth and expression of the progesterone-receptor using estrogen-sensitive-human breast cancer cells (MCF-7) at concentrations ranging from 10pM to 10μM. Cell-growth assays of the expression of progesterone-receptor mRNA by MCF-7 showed that diethylstilbestrol (positive control) and three chemicals, including bisphenol A, had strong estrogenic activity. Seven chemicals had weakly estrogenic activity. Another set of chemicals and phthalates had no estrogenic activity. We suggest that phenol rings are responsible for the estrogenic activity. Cell growth and expression of progesterone-receptor mRNA by MCF-7 cells in cultures containing both bisphenol A and the estrogen-receptor antagonist Tamoxifen was less than that of cultures containing BPA alone. These results suggest that BPA-enhanced cell growth is mediated by estrogen receptors in MCF-7 cells.
