Abstract : | 歯周病原細菌であるPorphyromonas gingivalisは, 多くの病原細菌と同様に細胞に侵入する事が報告されている1~4). 細胞内に侵入した病原細菌は宿主の免疫システムから逃れられるだけでなく, サイトカイン産生等を制御して病変を拡大する事が明らかにされている. 中でもインターロイキンー8(IL-8)は, 好中球走化因子として働く炎症性サイトカインであり, 細菌感染の初期防御因子として重要である. 歯周疾患においてもFusobacterium nucleatumやActinobacillus actinomycetemcomitansの刺激により歯肉上皮細胞(GEC)から産生が誘導されることが知られているが, P. gingivalisはGECからのIL-8産生を抑制する事により, 多型核白血球の浸潤を阻害し宿主の免疫応答に対抗していると考えられている5~8). 一方, 病原細菌の細胞内侵入には, 細菌由来分子に対する細胞側のレセプターを介した応答が必要であり, P. gingivalisの侵入には, 宿主細胞Ca2+の増加に伴うアクチン構造の再構築が必要である事が報告されている9). |

Abstract : | Interleukin-8 (IL-8) is a prointlammatory chemokine which attracts and activates neutrophils in innate immune system. Several periodontal bacteria are known to elicit IL-8 production from gingival epithelial cells (GEC), whereas Porphyromonas gingivalis infection fails to induce IL-8 or degrades its activity. In this study, we examined the activation of Mitogen-activated protein kinase (MAPK) pathways in GEC infected with P. gingivalis and the levels of IL-8 production were estimated. Uninfected GEC constitutively produced IL-8 and both ERK1/2 and MEK1/2 were activated. Exposure of GEC to F. nucleatum caused remarkable increase of IL-8 production in a dose-dependent manner. Furthermore, F. nucleatum markedly phosphorylated ERK1/2 in early incubation period. When GEC infected with F. nucleatum were exposed to U0126, specific inhibitor of MEK1/2, IL-8 level was significantly reduced. In contrast, P. gingivalis significantly reduced IL-8 activity in GEC and both ERK1/2 and MEK1/2 were transiently dephosphorylated. P. gingivalis strain YPP1 and YPP2, rgp and kgp mutant, respectively, showed comparable down-regulation of ERK1/2 activity in GEC to wild type strain 33277, however, had less ability to reduce IL-8 production. These results suggest that regulation of IL-8 production in GEC is associated with the phosphorylation of ERK1/2 and MEK1/2 and simultaneously P. gingivalis protease down-regulates IL-8 activity. |
