Abstract : 緒言 Porphyromonas gingivalis (以下, P. gingivalis)は成人性歯周炎や急速進行性歯周炎患者の歯周ポケット内から高頻度に分離され, 本菌に対する血清抗体価が有意に高い値を示すことから成人性歯周疾患の主たる病原細菌と考えられている1~5). Slotsらは歯周炎の様々な病型と特定のグラム陰性細菌の関連性を報告しており6), Whiteらは黒色色素産性桿菌の比率とポケット深さには正の相関があることを指摘している7). 偏性嫌気性菌である本菌の病原性に関しては, 歯周ポケットヘの定着に関与する線毛や血球凝集素, 細胞間基質を分解するコラゲナーゼやトリプシン様酵素, さらに免疫系細胞を活性化する内毒素や食細胞への抵抗因子となる莢膜などが挙げられる8). 感染症は, 病原微生物が特有の機構を介して粘膜や皮膚等の表面に付着し, 増殖を経て病巣を形成することによって発症する. 歯周病も歯肉溝内にP. gingivalisが付着することが発症の第一段階と考えられる.

Title : A Study of Relationship between Porphyromonas gingivalis 53K Fimbriae and Periodontitis

Abstract : We previously reported the existence of different kinds of fimbriae expressed by Porphyromonas gingivalis ATCC 33277. The properties of the 53K fimhriae of P. gingivalis 381 are different from those of the 67K fimbriae of ATCC 33277 as well as those of the 41K Fimbriae. Previously, we developed a host-vector system for P. gingivalis consisting of the host strain, YH522, a restriction-deficient derivative of SU60, and a plasmid vector, pYH420, capable of replicating stably in P. gingivalis. In this study, a fragment containing the 53K fimbrillin gene that encode the 53K fimbrillin of P. gingivalis 381 was subcloned in the vector pYH420, and the resulting recombinant plasmid was electroporated into restriction-deficient P. gingivalis YH522. The molecular mass of the recombinant fimbrillin from YH522 was evaluated by sodium dodecyl sulfate-polyacrylamide gel electrophoresis as 53K, which was identical to that of the native fimbrillin of P. gingivalis 381. The amino acid sequences of the 15 amino-terminal residues of the recombinant fimbrillin and the native fimbrillin of the strain 381 were identical. The involvement of P. gingivalis fimbriae in the invasion process and alveolar bone resorption in rats were examined. Adherence and invasion of P. gingivalis was assessed in human oral epithelial KB cells. We used a rat model to examine the role of the 53K fimbriae in alveolar bone loss by oral infection. The adherence and invasion levels of the transformant were higher than the wild-type strain. The bone loss of rats infected with the transformant was higher than that of those infected with wild-type strain. These data suggest that the 53K fimbrial protein play important roles in the pathogenesis of periodontal disease.
