Abstract : 緒言 歯根膜は発生学的には歯槽骨およびセメント質と同様に歯小嚢由来で,これらの石灰化組織に囲まれている1~4). 歯根膜線維芽細胞は骨系細胞の性質5~7)を持っているが,歯根膜は生理的に石灰化を起こさない. 歯根膜由来細胞を培養すると石灰化をするとの報告はある8~11). ここで培養された石灰化能を持った細胞は骨芽細胞に分化した細胞であって,歯根膜の線維芽細胞ではないと考えるのが妥当である. 歯根膜線維芽細胞はS100 calcium-binding protein familyの一つであるS100A4を産生12~14)し,S100A4が石灰化抑制機構に関わっているとの報告がある15). すなわちS100A4の発現を抑制すると骨芽細胞のマーカーのosteopontin(OPN)およびosteocalcin(OC)の発現が上昇し,また骨芽細胞に特異的な転写因子のRunx2/Cbfa1とOsterixの発現も上昇した. しかしこの報告15)は石灰化が阻害されたとの言及にはいたっていない. 一方,歯根膜に幹細胞が存在するとの報告は歯槽骨由来の骨髄からの未分化間葉系幹細胞が遊走した可能性を示唆している16~20).

Title : Human Periodontal Ligament Fibroblasts Inhibit Nodule Formation of Rat Bone Marrow Cells

Abstract : Periodontal ligament is a fibrous connective tissue located between two mineralized tissues of alveolar bone and cementum that remains unmineralized physiologically. Periodontal ligament contains mainly fibroblasts that have osteoblastic characters expressing bone-associated markers. The putative stem cells in periodontal ligament have been identified. It is not known that periodontal ligament cells possess regulatory mechanism to inhibit mineralization. The confluent rat bone marrow cells (RBMC) were co-cultured directly and indirectly with human periodontal ligament-derived fibroblast (HPLF) for 9 days. They were examined for the rates of cell growth, alkaline phosphatase (ALPase) activity, nodule formation and levels of mRNA for osteogenic regulatory proteins. RBMC were cultured in differentiation medium, which composed of α-minimun essential medium (α-MEM) supplemented with 10 nM dexamethasone (Dex) and β-glycerophosphate (β-GP). HPLF inhibited the nodule formation of RBMC that was stimulated by the differentiation medium. The nodule formation of RBMC cultured with insert HPLF or protein components of conditioned medium of HPLF (CM) were decreased dramatically. The nodule formation of RBMC contacted and cultured with HPLF was almost undetectable. RBMC contacted with HPLF decreased the expression of Runx2 (Cbfal) and osterix mRNAs that regulate the osteogenic differentiation as transcription factors. To maintain the periodontal ligament, the mesenchymal stem cells were necessary. These results suggested that cells in periodontal ligament regulated voluntarily the inhibition of mineralization.
