Abstract : 「緒言」歯周組織は歯を支持する組織であり, 歯根膜, 歯槽骨, セメント質から構成されている. 特に歯根膜は腱靭帯と構造の類似した強固な結合組織で構成されており, 咬合力に対する緩衝作用を有するばかりでなく, 咀嚼刺激を中枢に伝える感覚受容器の機能も有する1). しかし, 慢性炎症性疾患である歯周病に罹患すると歯根膜は非可逆性の崩壊を受け, 重度に進行したケースでは機能障害に陥る. そのため, 歯根膜の再生を目的とした歯周組織再生療法が開発され, Guided Tissue Regeneration(GTR法)2,3)やエナメルマトリックスタンパク質(エムドゲイン)4,5)により, 歯根膜は再生可能であることが報告されてきた. しかし, 歯根膜再生のメカニズムが十分に理解されていないため, 未だ歯周病の創薬ターゲットは同定されていないのが現状である. この問題を解決するには, 歯根膜発生機構を理解することが重要である.

Title : Function analysis of scleraxis during periodontal ligament formation

Abstract : Scleraxis (Scx), a bHLH transcription factor, is involved in the compartmentalization of tendon/ligament primordia or differentiation of tendon progenitor during tendon/ligament development. Recently, we have demonstrated that Scx may play an important role during periodontal ligament (PDL) formation. We report that Scx negatively regulates the mineralization in the PDL. In situ hybridization analysis revealed that Scx was expressed in mature PDL, but not detected in the dental follicle which include progenitor for PDL during tooth germ development. To investigate Scx function in PDL, Scx was transduced into MDFE6EGFP, an immortalized mouse dental follicle cells (MDFScx) with retrovirus transfer. MDFScx had capacity to generate PDL-like tissue that showed well organized cell alignment and sheet-like appearance upon implantation into immunodeficiency mice. Gene expression analysis revealed that overexpression of Scx induced down-regulation of osteoblastic phenotype-related genes such as bone sialoprotein, Osteocalcin and Osterix. In addition, mineralization potential of MDFScx was down regulated compared with those of MDFE6EGFP. These results suggest that Scx act as a negative regulator of mineralization for maintaining of periodontal ligament compartment.
