Abstract : 「緒言」 ヒト顎下神経節は頸部の顎下三角において顎下腺主導管に接して存在し, 唾液の分泌調節に関与する1). またラット顎下神経節は小型で唾液腺主導管に沿い顎下腺の門付近に主として存在し, 結合組織性の被膜に囲まれる2, 3). 顎下神経節は顎下腺および舌下腺の唾液分泌量および唾液成分を調節する交感神経と副交感神経の神経節であるとされてきた. しかし, 顎下神経節のニューロン間における神経回路網の存在について詳細な研究は行われていない. 顎下神経節に分布する交感神経性の神経線維は上頸神経節ニューロンに由来する節後線維であり, また副交感神経性の神経線維は延髄の上唾液核に由来する節前線維であり, コリン作動性である. これに対して近年, 顎下神経節に分布する神経線維について非コリン作動性を示唆する種々の神経活性物質が顎下神経節に示されてきた2). すなわち, vasoactive intestinal polypeptide(VIP), enkephaline(ENK)4, 5), neuropeptide Y(NPY)6), substance P(SP), calcitonin gene related peptide(CGRP), serotonin(5-HT)である.

Title : Immunohistochemical Study of Effect of Antidepressant Drugs on Down-regulation of Serotonin in the Rat Submandibular Ganglion Neurons.

Abstract : A possible cause of hydrodipsia is the administration of drugs that act on the central nervous system, particularly selective serotonin reuptake inhibitors (SSRI) prescribed as antidepressants. Although the detailed mechanism of action of SSRI has not yet been clarified, such form of hydrodipsia has been confirmed in dental practice. Secretion of saliva from the acinar cells of the submandibular and sublingual glands is regulated by the submandibular ganglion, one of the parasympathetic ganglia in the head and neck region. In this study, down-regulation of serotonin (5-HT) receptors in the neurons of the submandibular ganglion was investigated immunohistochemically in rats following administration of SSRI (paroxetine hydrochloride hydrate). Double-immunohistochemical staining of 5-HT and S-100 protein post administration, 5-HT immunoreactivity was not observed in any ganglion cells. In addition, no 5-HT immunoreactivity was observed in the satellite cells surrounding the ganglion neurons at this time. Serotonin immunoreactivity was not observed in the ganglion neurons at 3 hours post administration, but was observed at 12 hours post administration. These results suggest that the SSRI had a direct effect on the submandibular ganglion neurons by upregulating their synthesis of 5-HT and thereby modifying the secretion of saliva through the regulation of the epithelial cells of the main duct of the salivary glands.
