Abstract : 「緒言」ミトコンドリアDNA(mtDNA)は細胞質に存在する小器官であり, 環状の二重鎖構造をしている. 約1,000塩基で構成されるD-loop領域(非コード領域)には, 非常に多くの塩基の欠失, 置換や挿入による変異が観察され, 高度多型領域(Hyper Variable Region (HVR))1-4)と呼ばれている. HVRにはHVR1, HVR2, HVR3の3つの領域が報告されており, これらの領域の塩基配列の多型性を利用した個人識別が法医学において広く用いられている5-11). また, コード領域において認められる点変異は, 疾病と相関関係があるとされている12,13). さらに, 核DNAが哺乳動物の細胞1個にひとつ存在するのに対して, mtDNAは細胞1個中に数千個存在することから, 非常に低分子化した試料や極微量の試料からでもPCR増幅を利用して解析が可能であることが多数報告されている14-20).

Title : Availability of Cloning Methods in DNA Testing in Criminal Investigation

Abstract : The polymerase chain reaction (PCR) and direct DNA sequencing have enabled rapid and reliable characterization of specific highly polymorphic DNA sequences from different samples. Human mitochondrial DNA (mtDNA), which has an extremely high copy number, possesses such a highly polymorphic region. Even if human chromosomal DNA is degraded, the hypervariable D-loop region of mtDNA can be amplified by PCR and cloned for sequencing to identify individuals. We have established a new method for independently sequencing each clone derived from PCR products, and applied it to contaminated samples that are occasionally encountered in various fields of forensic medicine. In this study, individual identity was confirmed from trace samples using this method. HVR1 and HVR2 in the D-loop region of the target mtDNA extracted from the trace samples were amplified by PCR. After confirming the amplification, the DNA of HVR1 region was cloned, amplified and then directly sequenced. The sequences obtained are consistent with each other, demonstrating that each sample was derived from the same individual. These results suggest that the direct sequencing of the HVR1 and HVR2 in the D-loop region of the target mtDNA and cloning is applicable to the practice of forensic medicine, and that the method allows faster and more accurate identification of individuals.
