Abstract : 「1. 研究背景」唾液腺は消化だけでなく, 様々な役割を担っている. 過去の唾液腺の研究をひも解くと, 典型的な外分泌腺であるにもかかわらず, 古くから内分泌腺的な機能を含めて多くの付加的な機能についての研究がなされてきた. 特に本邦においてはウシ耳下腺から抽出精製されて, 血中カルシウム濃度の低下や石灰化促進作用を有するといわれた「パロチン」が有名である1). また, 「細胞成長因子」の最初の発見である神経成長因子(Nerve Growth Factor:NGF)とそれに引き続いて上皮成長因子(Epidermal Growth Factor:EGF)の両方がマウス顎下腺から抽出されて, 唾液腺の新たな機能が注目されるに至った. 2つの細胞成長因子の発見後, さらに数多くの増殖因子が発見されている. また, げっ歯類の唾液腺で産生される物質の一部が血中濃度に影響していることが知られている.

Title : The preventive medicine research in stress-induced diseases suppression of stress-related salivary gland protein BDNF - Toward the creation of the new functions in salivary gland -

Abstract : Brain-derived neurotrophic factor (BDNF) belongs to the neurotrophin family, which includes nerve growth factor. A high level of BDNF expression has been observed in the hippocampus. BDNF plays an important role in the survival, differentiation, maintenance, and protection of neural cells from insult. The results of our study demonstrated expression of BDNF in the submandibular gland in response to stress, with maximal elevation occurring following a 60-min period of immobilization-induced stress. The submandibular glands were the main source of plasma BDNF under this stress condition. In contrast, only the adrenal glands showed significantly increased levels of BDNF receptor TrkB mRNA after 60 min of stress. Exposure of PC12 cells derived from the medulla of the adrenal gland to BDNF induced release of catecholamine. Thus, BDNF-TrkB interactions may modulate catecholamine release from adrenal chromaffin cells under acute stress conditions. The results of this study suggest that the salivary gland exerts hitherto unknown effects on the functioning of the body as a whole and that it plays an important role in maintaining health. This indicates that the value of this organ will have to be reevaluated in terms of systemic health, and that this awareness will lead to the development of new tests and treatments in the field of dentistry.
